We are looking for a motivated and proficient post doc in a new project where we characterize plant polyketide reductases (PKRs) using enzymatic and genomic approaches.
A vast array of plant secondary metabolites are synthesized through the acetate-malonate pathway, where acetate units provided by malonyl-CoA are concatenated into a polyketide intermediate which may fold in a number of ways to produce, e.g., new aromatic ring systems. The best known example of these reactions is the formation of chalcone, the first precursor of the flavonoid branch of phenylpropanoids. Chalcone synthase together with other plant polyketide synthases form a super family with a common origin in evolution but wide variation in starter substrate selection, number of iteration steps and final folding of the product. In analogy to fatty acid biosynthesis, but in a highly selective manner, the growing polyketide chain may also be reduced during its synthesis, leading to further increased chemical variation. The associated PKR is known only for chalcone synthase, but reductive steps are needed for biosynthesis of, e.g., the raspberry ketone (raspberry), the glucosidic lactones gerberin and parasorboside (gerbera), the alkaloid coniine (poison hemlock) and the naphtoquinone plumbagin (sundews). The project is a joint project between University of Helsinki and VTT. Our specific objectives are to identify and isolate genes encoding PKRs in several plants, to heterologously express the PKR genes and isolate the encoded enzymes and to functionally characterize the enzymes in combination with the relevant PKS and analyze the products. Genomic and transcriptomic resources exist for gerbera and will be developed for the other plant species. The Academy of Finland funded postdoctoral position is for 2011-2014. Suitable candidates should have an interest in plant secondary metabolism and experience in the areas of molecular biology, protein purification, enzymology and bioinformatics.
To apply, e-mail your application, CV, and contact information of two referees to teemu.teeri- at-helsinki.fi
Deadline for applications is January 15, 2011. Additional information about the research group: teemu.teeri-at-helsinki.fi http://www.mm.helsinki.fi/mmsbl/english/research/Gerberalab/SecMetab.html

A four-year post-doctoral position is available in the research group of Jukka Finne at the Department of Biochemistry and Biotechnology, University of Helsinki, Finland. The project deals with the identification of regulatory factors of polysialic acid, an important developmental cell surface antigen also associated with the malignant behaviour of cells, using novel insertion mutagenesis technology. The position is to be filled as soon as possible. Applications will be considered until a suitable candidate has been found. (9.12.2010)
