Please visit the course homepage for more information about the course: www.biostatepi.org 20-26/12/2010 INFO 3 We are looking for a motivated and proficient post doc in a new project where we characterize plant polyketide reductases (PKRs) using enzymatic and genomic approaches. A vast array of plant secondary metabolites are synthesized through the acetate-malonate pathway, where acetate units provided by malonyl-CoA are concatenated into a polyketide intermediate which may fold in a number of ways to produce, e.g., new aromatic ring systems. The best known example of these reactions is the formation of chalcone, the first precursor of the flavonoid branch of phenylpropanoids. Chalcone synthase together with other plant polyketide synthases form a super family with a common origin in evolution but wide variation in starter substrate selection, number of iteration steps and final folding of the product. In analogy to fatty acid biosynthesis, but in a highly selective manner, the growing polyketide chain may also be reduced during its synthesis, leading to further increased chemical variation. The associated PKR is known only for chalcone synthase, but reductive steps are needed for biosynthesis of, e.g., the raspberry ketone (raspberry), the glucosidic lactones gerberin and parasorboside (gerbera), the alkaloid coniine (poison hemlock) and the naphtoquinone plumbagin (sundews). The project is a joint project between University of Helsinki and VTT. Our specific objectives are to identify and isolate genes encoding PKRs in several plants, to heterologously express the PKR genes and isolate the encoded enzymes and to functionally characterize the enzymes in combination with the relevant PKS and analyze the products. Genomic and transcriptomic resources exist for gerbera and will be developed for the other plant species. The Academy of Finland funded postdoctoral position is for 2011-2014. Suitable candidates should have an interest in plant secondary metabolism and experience in the areas of molecular biology, protein purification, enzymology and bioinformatics. Deadline for applications is 15 January 2011. 13-19/12/2010 INFO 3 Postdoctoral position in the lignin biosynthesis laboratory, Department of Biosciences, University of Helsinki. We are looking for a motivated and proficient post doc in a project where we characterize monolignol transport proteins at the plasma membrane using biochemical and bioinformatics approaches mainly with Norway spruce, while Arabidopsis and hybrid poplar are used when needed. As the means by which monolignols are transported from the protoplasts into the apoplastic space is not known yet, we are interested in all possible transport methods including ABC-type plasma membrane transporters and diffusion based on the hydrophobic- hydrophilic properties of the molecules to be transported. The work entails isolation of plasma membranes of Norway spruce and hybrid poplar and analysis of their transport proteins and characterization of the putative monolignol transporters. As diffusion of monolignols is also a possibility, this will be analyzed using the isolated plasma membrane vesicles. The work is in progress and many of the methods to be used have been optimized for the particular plant material in this project. The task belongs to a collaborative research project, FuncWood 3, between Helsinki University and the Finnish Forest Research Institute, and is funded by the Finnish Forest Cluster EffTech-programme. The postdoctoral position is available for the period 1st March 2011 till 30th June, 2013. The government’s Cabinet Finance Committee has approved a decree by the Ministry of the Interior to raise the processing fee for a foreign national’s first Finnish residence permit from 225 euros to 350 euros, and for a first residence permit for study purposes from 125 euros to 250 euros, to take effect at the beginning of next year. Biometric identifiers will be adopted in residence permits next year. The introduction of new smart-card residence permits that include fingerprints and photo ID will lift costs in comparison to the current document, which is no more than a one-euro sticker inside the permit-bearer’s passport. The price hike for student-purpose residence permits is also justified on the grounds that establishing the authenticity of foreign educational diplomas requires extra man hours. Quota refugees and foreign nationals seeking asylum are not charged any fees. The fee for citizenship applications will rise to 440 euros.
